The Edvotek® Blue/White Cloning of a DNA Fragment experiment provides students with a comprehensive introduction to recombinant DNA technology and one of the most widely used screening methods in molecular biology. This hands‑on laboratory activity demonstrates how insertion of a DNA fragment into a plasmid can disrupt β‑galactosidase production, allowing recombinant colonies to be easily identified.
In this experiment, a provided DNA fragment is ligated into a linearised pUC‑based plasmid using T4 DNA ligase. When the DNA is successfully inserted into the plasmid’s polylinker region, β‑galactosidase activity is interrupted. Following transformation of competent E. coli cells, students plate the bacteria on selective media containing X‑Gal and antibiotics. Recombinant cells form white colonies, while non‑recombinant cells form blue colonies.
Students then extend their investigation by performing a β‑galactosidase activity assay on both blue and white colonies, reinforcing the link between genotype and phenotype. This experiment can be completed as a full workflow or divided into three flexible modules: plasmid ligation, bacterial transformation, and enzymatic assay.
